Existing vaccines have several limitations. Research new vaccine platforms will enable the development of more effective strategies of disease prevention.</p> <p><bold>Materials and methods.</bold> The constructs for producing vaccine preparations were obtained using the methods described earlier. Study of the effectiveness of candidate vaccines based on chimeric yellow fever virus, adenovirus serotype 25 and mRNA encoding a fragment of the tick-borne encephalitis viral polyprotein corresponding to the PrM and E regions, immunogenicity and protective efficacy was performed using BALB/c mice. The neutralizing activity assessment of animal sera and mice challenge were performed using the tick-borne encephalitis virus strain Sofjin-Chumakov.</p> <p><bold>Results.</bold> It was established that immunization of animals with mRNA encoding the PrM and E regions of the viral polyprotein of tick-borne encephalitis virus resulted in the induction of a higher level of neutralizing antibodies compared to the other vaccine platforms studied in the immunization schedule used. In addition, this platform provided 100% protection of animals from the lethal infection caused by the tick-borne encephalitis virus.</p> <p><bold>Conclusion.</bold> The study demonstrated that delivery of the nucleotide sequence encoding the PrM and E regions of the tick-borne encephalitis virus polyprotein within mRNA elicits a pronounced humoral immune response during animal immunization. These results demonstrate the fundamental feasibility of effectively expressing the same tick-borne encephalitis virus antigen in various vaccine platforms, each with its own advantages and limitations.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение.</bold> К настоящему времени не решена проблема распространения заболеваемости вирусом клещевого энцефалита (<italic>Orthoflavivirus encephalitidis</italic>). Имеющиеся вакцины имеют ряд недостатков. Изучение новых вакцинных платформ позволит разработать более эффективные вакцины для профилактики заболевания.</p> <p><bold>Цель исследования.</bold> Изучить эффективность кандидатных вакцин на основе химерного вируса желтой лихорадки, аденовируса обезьян 25-го серотипа и матричной рибонуклеиновой кислоты (мРНК), кодирующих фрагмент вирусного полипротеина клещевого энцефалита, который соответствует областям PrM и E.</p> <p><bold>Материалы и методы. </bold>Конструкции для вакцинных препаратов получали по описанным ранее методикам. Изучение иммуногенности и протективности вакцин проводили с использованием мышей линии BALB/c. Оценку нейтрализующей активности сывороток животных, а также инфицирование мышей проводили с использованием вируса клещевого энцефалита штамм «Софьин-Чумаков».</p> <p><bold>Результаты. </bold>Установлено, что иммунизация животных мРНК, кодирующей области PrM и E вирусного полипротеина вируса клещевого энцефалита, приводила к индукции более высокого уровня нейтрализующих антител по сравнению с остальными исследуемыми вакцинными платформами в используемой схеме иммунизации. Кроме того, данная платформа обеспечивала 100% защиту животных от летальной инфекции, вызываемой вирусом клещевого энцефалита.</p> <p><bold>Заключение.</bold> Проведенное исследование показало, что доставка нуклеотидной последовательности, кодирующей области PrM и E вирусного полипротеина вируса клещевого энцефалита, в составе мРНК вызывает выраженный гуморальный иммунный ответ при иммунизации животных.
