All infected patients were divided into 3 groups, depending on disease severity assessment, which was appreciated by NEWS2. Another group consisted of participants, who had asymptomatic infection in the past. Determination of SNPs of the genes <italic>FGB</italic> (-455 G/A), <italic>FII</italic> (20210 G/A), <italic>FV </italic>(1691 G/A), <italic>FVII</italic> (10976 G/A), <italic>FXIIIA1</italic> (103 G/T), <italic>ITGA2</italic> (807 C/T), <italic>ITGB3</italic> (1565 T/C), <italic>SERPINE1</italic> (-675 5G/4G) were performed by PCR using the “Genetics of Hemostasis” kit (“DNA-Technology”, Russia).</p> <p><bold>Results. </bold>In analyzed SNPs, no significant differences were detected between the group of infected patients and healthy participants. But significant association was revealed in gene <italic>SERPINE1</italic> (-675 5G/4G), when patient groups, differing in the disease severity, were analyzed relative to the group of participants with asymptomatic infection (p=0.0381; p=0 .0066; p=0.0009). It was found, that as COVID-19 severity scores increased, the proportion of 5G allele of gene <italic>SERPINE1</italic> decreased, and the proportion of the 4G allele increased (p=0.005; p=0.009; p=0.0005). Similar processes were observed for genotypes 5G/5G and 4G/4G.</p> <p><bold>Discussion.</bold> The gene <italic>SERPINE1</italic> (-675 5G/4G) is associated with the severity of COVID-19.</p> <p><bold>Conclusion.</bold> For the first time, it was discovered that 5G/5G genotype of gene <italic>SERPINE1</italic> (-675 5G/4G) can be a marker of a milder course of COVID-19, and the 4G/4G genotype as a more severe one.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение.</bold> COVID-19 характеризуется разнообразным течением: от бессимптомного до тяжелого с летальным исходом. Комплекс неблагоприятных факторов инфицированного организма во многом определяет течение и исход болезни.</p> <p><bold>Цель работы – </bold>выявить возможные ассоциации полиморфных генов системы гемостаза с течением COVID-19.</p> <p><bold>Материалы и методы. </bold>ДНК выделяли из крови инфицированных пациентов (n=117) и участников группы сравнения (n=104). Все инфицированные были поделены на 3 группы в зависимости от тяжести состояния, которую оценивали по NEWS2. Еще одну группу составили участники, перенесшие инфекцию бессимптомно. Определение однонуклеотидного полиморфизма (ОНП) генов <italic>FGB </italic>(-455 G/A), <italic>FII</italic> (20210 G/A), <italic>FV</italic> (1691 G/A), <italic>FVII</italic> (10976 G/A), <italic>FXIIIA1</italic> (103 G/T), <italic>ITGA2</italic> (807 C/T), <italic>ITGB3</italic> (1565 T/C), <italic>SERPINE1</italic> (-675 5G/4G) проводили методом ПЦР, используя набор «Генетика гемостаза» («ДНК-Технология», Россия).</p> <p><bold>Результаты. </bold>Достоверных различий в аллельных вариантах анализируемых генов между группой всех инфицированных и здоровыми участниками не выявлено. Но анализ групп пациентов, различающихся по тяжести COVID-19, относительно группы участников, бессимптомно перенесших инфекцию, показал, что в гене <italic>SERPINE1 </italic>(-675 5G/4G) обнаружена достоверная ассоциация ОНП с течением инфекции (р=0,0381; р=0,0066; р=0,0009). Установлено, что по мере увеличения баллов тяжести COVID-19 доля аллеля 5G гена <italic>SERPINE1</italic> (-675 5G/4G) снижалась, а доля аллеля 4G увеличивалась (р=0,005; р=0,009; р=0,0005). Аналогичные процессы наблюдались для генотипов 5G/5G и 4G/4G.</p> <p><bold>Обсуждение</bold>. Анализ вклада ОНП генов системы гемостаза показал, что фибринолитическое звено ассоциировано с тяжестью COVID-19. Полиморфизм гена этого звена <italic>SERPINE1</italic> (-675 5G/4G) имел достоверную связь с течением COVID-19.</p> <p><bold>Заключение.</bold> Впервые обнаружено, что ОНП гена <italic>SERPINE1 </italic>(-675 5G/4G) ассоциирован с течением COVID-19.
